The major isozyme of rat cardiac glutathione transferases. Its correspondence to hepatic transferase X.

1986

1. A major isozyme of rat heart glutathione transferase was purified to homogeneity by Sephadex G-200 gel filtration, ammonium sulfate precipitation, CM-cellulose chromatography and affinity chromatography on S-hexylglutathione-linked Sepharose 6B. 2. The purified isozyme was a dimer with an apparent relative molecular mass of 50000 composed of two Yb-size subunits (Mr= 26 500). The isozyme is immunologically related to rat liver glutathione transferase X and 3–3, especially closely to transferase X, and no immunological cross-reactivity with subunits 1 and 2 of hepatic glutathione transferases was observed. The isoelectric point (pI = 6.9) of the isozyme was identical with and the substrat…

Assessing the genetic diversity ofCentaurea parlatorisgroup (sect.Dissectae, Compositae) in Sicily using isozymes

2011

Abstract The Centaurea parlatoris group belongs to sect. Dissectae and is one of the most taxonomically critical groups in Sicily. The taxa included in it inhabit dry slopes, pastures and rocky places. Some of them are narrow endemics to Sicily, and others to Italy. The great morphological variability at the intrapopulation level has not permitted the creation of an adequate taxonomic scheme. The recent proposal of two new species from Sicily confirms the insufficient knowledge of the taxonomic diversity. This study involves eight Sicilian populations of the C. parlatoris group. Seven loci from nine enzyme systems [isocitrate dehydrogenase (IDH), malate dehydrogenase (MDH), 6-phosphoglucona…

Genetic diversity in theCentaurea cinerariagroup (Compositae) in Sicily using isozymes

2006

Abstract The genetic diversity of seven taxa endemic to Sicily, C. cineraria, C. busambarensis, C. ucriae subsp. ucriae, C. ucriae subsp. umbrosa, C. todari, C. erycina and C. saccensis, from 11 localities was investigated using isozymes. Eight loci from five enzyme systems (IDH, MDH, PGD, PGM and PGI) were examined. A total of 19 alleles were identified, some rare and two of them exclusive to different populations. The allelic frequencies and the genetic variability values for each population were calculated. On the whole, the genetic diversity, i.e., average polymorphism (P) = 0.41, average number of alleles per locus (A) = 1.75, Nei's gene diversity (H) = 0.18, is moderate, with the high…

Genetic variation patterns in some groups of Centaurea (Asteraceae) in Italy

2012

In the current work the data of the isozyme analysis of all the twenty studied Italian Centaurea taxa were compared. They belong to Centaurea parlatoris, C. cineraria, C. tenorei and C. jacea groups. The following parameters of the genetic variability were considered: the total number of the alleles, the mean number of alleles per locus (A), the mean percentage of polymorphic loci (P95), the expected heterozygosity (He) that is a measure of the intra-population diversity, the Wright’s fixation index (F) and inbreeding coefficient (FIS) that are measures for the deviation from the Hardy-Weinberg equilibrium. The present study contributes to insight some aspects of the genetic diversity relat…

Polyoxypregnanes as safe, potent, and specific ABCB1-inhibitory pro-drugs to overcome multidrug resistance in cancer chemotherapy in vitro and in vivo

2021

Multidrug resistance (MDR) mediated by ATP binding cassette subfamily B member 1 (ABCB1) is significantly hindering effective cancer chemotherapy. However, currently, no ABCB1-inhibitory drugs have been approved to treat MDR cancer clinically, mainly due to the inhibitor specificity, toxicity, and drug interactions. Here, we reported that three polyoxypregnanes (POPs) as the most abundant constituents of Marsdenia tenacissima (M. tenacissima) were novel ABCB1-modulatory pro-drugs, which underwent intestinal microbiota-mediated biotransformation in vivo to generate active metabolites. The metabolites at non-toxic concentrations restored chemosensitivity in ABCB1-overexpressing cancer cells v…

Isolation and biodiversity inCytisus villosusPourret (Fabaceae, Genisteae): enzyme polymorphism in disjunct populations

1997

ABSTRACT The genetic diversity of isolated populations of Cytisus villosus has been studied by means of enzyme polymorphism analysis. Two types of isolated populations were studied: “terrestrial islands” in Sicily, and “true islands” in the Aeolian archipelago. In the populations of “true islands” the number of alleles and the heterozygosity are lower than in “terrestrial islands”. Isolation amongst Sicilian populations seems to be more recent than isolation of the Aeolian populations, and may be attributed to climatic changes which occurred during the Holocene and/or to human activities. The disjunction of the Aeolian populations seems much more recent than the origin of the isles themselv…

Isozyme analysis of genetic diversity in wild Sicilian populations of Brassica sect. Brassica in view of genetic resources management

2004

In Sicily and in the small surrounding islands the section Brassica of the genus Brassica comprises five species, B. insularis Moris, B. incana Ten., B. macrocarpa Guss., B. rupestris Raf. and B. villosa Biv. These taxa represent a genetic resource as relatives of kale crops but some populations are endangered or threatened, thus isozyme analyses were performed to assess the genetic diversity degree at population and species levels in order to assist the design of conservation management programs. Eleven loci from five enzyme systems (aconitase, leucine aminopeptidase, 6-phosphogluconate dehydrogenase, phosphoglucoisomerase phosphoglucomutase) were analyzed in sixteen natural population (fi…
